Can I use POST /api/v1/gst/calculate to file GST or verify a GSTIN against GSTN?

No. POST /api/v1/gst/calculate returns exclusive or inclusive GST with a CGST/SGST or IGST split from the amount and rate you send. It is not GST filing, e-invoice, place-of-supply determination for every legal case, or GSTIN verification against GSTN. Results are not tax advice. Use a Developer Console TEST key on mantechstudio.in/api/v1.
